 ESP Drifter and Hawaii Cruise

•  Chris/Julie's preliminary work shows that waste volume allows only 40 assays.  Thought is to perform 40 WCR's and 17 PCR's.
  • We could change MFB Final Purges to Final Purges to save on waste space.
  • Want 2L volume for each sample.  Need to know how long for power-budget calculations.
  • Julie to test both the above issues,
• Plan is to do DeLong's stuff first (2-3 days of WCR every 2 hours), and intersperse the PCR within that.  DeLong may lose 2-3 WCR's with this scheme.
• Chris S. suggests dropping it near a front so it will eventually cross.
• Julie has filters from the latest ALoha cruise.  She'll use these to create lysate and see how well her p/p work.
• Using MOE for this deployment.  Currently, CS is rusted, and waste container seems to be leaking.  Roman to work on before leaving Saturday.
• Schedule:
  • Cruise is Sep 6-21.
  • Will need 1 week prior to MOB to run dockside standards.
  • Will ship instrument in refrigerated van, reagents loaded and closed, but not final seal.
  • Ship on ~ Aug 19, depending on transit time.

Array printer

•  alignment still needed.  Hopefully finished by PM today.  Will print tomorrow.  Chris says she doesn't need arrays, but current print needs testing to make sure.  Roman operating as if more are needed.

• Chris P. and Bill have put much work into a straw man schedule. It is HERE.
• We will not purchase an optode and refit our Seabird CTD.  Too much $$. ($5K for optode, $3K to Seabird for firmware changes, plus cables)
• Do we use the Clark?  TBD.  Must check with company about H2S effects on electrode.  Are effects reversible?  Bill looking into.

HAB PCR

• Elif working on getting probes for SHA bugs.
